Abstract目的 探讨局部注射C型钠尿肽(C-type natriuretic peptide,CNP)对于大鼠骨折的愈合是否具有促进作用.方法 36只12周龄雄性SD大鼠,建立大鼠股骨干骨折模型,随机分为3组,每组12只.组1大鼠在断骨后,在骨折处局部注射溶于1 ml凝胶中的25 μg CNP;组2大鼠在断骨后,在骨折处局部注射凝胶1 ml;组3大鼠在断骨后,在骨折处局部注射医用生理盐水1 ml.在建模后每周对大鼠进行X线扫描检测.第3周时,随机从每组中各选取7只大鼠,在接受X线扫描后处死,股骨接受Micro-CT扫描检测.每组剩余的5只大鼠在第4周接受X线扫描后处死,股骨接受Micro-CT扫描检测.收集建模后第3、4周股骨及骨折部位样本进行生物力学检测.结果 X线显示组1在术后第3、4周均出现大鼠骨折部位骨桥的形成,组2、3所有大鼠直至实验结束也未出现骨折端有骨桥形成.Micro-CT结果显示术后第3周组1骨折端骨痂体积为(187.655±62.864)mm3,高于组2(137.703±52.592)mm3和组3(176.493±75.193)mm3,但无统计学差异(F=1.167,P=0.334);术后第4周组1、2、3的骨折端骨痂体积分别为(242.853±139.252)、(242.460±64.844)、(226.274±77.418)mm3,3组比较无统计学差异(F=0.045,P=0.956).组1第3周时4只大鼠达到骨折愈合标准,组2、3未出现骨折愈合大鼠;组1第 4周时4只大鼠达到骨折愈合标准,组2、3未出现骨折愈合大鼠.生物力学检测结果显示第3周时,组1中能够完成生物力学检测的大鼠为4只,组2为0只,组3为1只;第4周时,组1中能够完成生物力学检测的大鼠为4只,组2为0只,组3为0只.结论 在骨折局部注射CNP对SD大鼠骨折的愈合有一定的促进作用.
Objective To evaluate the promoting effect of C-type natriuretic peptide (CNP) on facture healing in a rat model.Methods A total of 36 Sprague-Dawley (SD) male rats were employed for induction of femoral fracture model in three randomly divided groups (12 for each group).Rats in Group 1 received local injection of 1 ml gel containing 25 μg of CNP, rats in Group 2 were treated with local injection of 1 ml of gel, and rats in Group 3 were provided with 1 ml of saline solution at the same position.The X-ray was performed for each group after fracture at each week (0, 1st, 2nd, 3rd, and 4th weeks).Furthermore, legs of 7 rats in each group were harvested in the 3rd week and the remaining 5 rats in each group were obtained in the 4th week.The progression of fracture healing of each specimen was assessed by micro-CT.Mechanical tests of the femora and fractures were carried out at the 3rd and 4th weeks.Results The X-ray showed that some rats of the Group 1 appeared the formation of bone bridge at the site of fracture at 3rd and 4th weeks after surgery, while the Group 2 and 3 did not appear bone bridge at the end of experiment in all rats.The results of micro-CT showed that the volume of callus in the Group 1 at the fracture site at 3rd week was (187.655±62.864) mm3, which was higher than that in the Group 2 (137.703±52.592) mm3 and Group 3 (176.493±75.193) mm3, but the differences were not statistically significant (F=1.167, P=0.334).At the 4th week, the volumes of callus in the Group 1, 2, and 3 were (242.853±139.252) mm3, (242.460±64.844) mm3, and (226.274±77.418) mm3, respectively, and there were no significant differences among the 3 groups (F=0.045, P=0.956).Four rats in the Group 1 reached the standard of fracture healing at the 3rd week, but none in the Group 2 or Group 3.At the 4th week, there were another 4 rats reaching the standard of fracture healing in the Group 1, but none in the Group 2 or 3.The biomechanical test results showed that 4 rats in the Group 1 completed the biomechanical test at the 3rd week, none in the Group 2, and 1 rat in the Group 3.At the 4th week, 4 rats completed the biomechanical test in the Group 1 but none in the Group 2 or Group 3.Conclusion Treatment with local CNP injection accelerates fracture healing in a SD rat model.
