Abstract目的:探讨NDRG 2基因在结直肠癌组织中mRNA的表达情况及其与临床病理特征之间的关系。方法:通过实时定量PCR检测68例结直肠癌组织及其配对癌旁组织中NDRG 2 mRNA的表达情况,同时完成5a随访,进而分析病人临床病理特征与术后生存率之间的关系。结果:(1) NDRG 2在68例结直肠癌组织中的mRNA表达量明显低于其配对癌旁组织(P<0.01),差异有统计学意义;(2)TNM分期早(TNM 1)的结肠癌组NDRG 2 mRNA的表达量高于分期晚(TNM 2/3/4)的结肠癌组(T1 VS. T2+3+4,P=0.04);而其余临床病理特征对于结直肠癌组NDRG 2 mRNA表达量的影响无显著差异性(P>0.05);(3)NDRG 2表达水平与结直肠癌病人生存率未见明显相关性(P>0.05)。结论:NDRG 2基因参与结直肠癌的发生发展过程,但具体机制尚未明确。NDRG 2在核酸层面的表达与部位、分化程度、浸润深度、Dukes分期、淋巴结侵犯、术前干预、复发及转移均无明显关系,仅与TNM分期有关,这为进一步探讨结直肠癌的分子诊断及治疗提供理论依据。
Objective:To explore the mRNA expression of NDRG 2 which is considered as candidate tumor - suppressor genes in colorectal carcinoma and the relationship with clinical pathological features. Methods:NDRG 2 mRNA levels were examined by real-time quantitative PCR in a cohort of 68 human colorectal carcinomas and 68 paired normal tissues. The correlation between NDRG 2 mRNA expression with clinicopathological features and survival followed up for 5 years after surgery was analyzed. Results:(1) NDRG 2 mRNA levels were found lower in tumor samples than in the normal tissues(P<0. 01);(2)The mRNA expression level of NDRG 2 was higher in patients with TNM 1 compared with TNM 2,3 and 4(T 2,3 and 4 compared to TNM 1 P=0. 04),but there was no significant differences of NDRG 2 mRNA expression between different groups of location differentiation invation Dukes-staging lymph node metastasis preoperation intervention, recurrence and distant metastasis( P>0. 05 );( 3 ) The expression levels of NDRG 2 were no statistical significance with patients survival. Conclusion:NDRG 2 plays an important part in the progress of colorectal tumor de-velopment, invasion or metastasis with unclear mechanism. NDRG 2 transcript levels revealed significant differences between varying TNM staging, which provides the guiding for the clinical diagnosis and prognosis in future.
